Format
Each kit is presented as a 96-well plate with one SmartBead (formulated with ~6,000 UltraCodes of six types) in each well. All buffers and detection reagents are provided. Each kit is suitable for determination of 32 samples in singlicate, together with positive and negative control sera.
Antigens
The kit uses native wheat gliadin purified from natural sources and recombinant human tissue transglutaminase (tTG) purified from a bacterial expression system. The assay also includes control Qmax zero microparticles, and microparticles coated with anti-human IgA for the total IgA assay.
Summary of Procedure
Diluted serum, from primary tubes, is added to the 96-well plate by the SmartStation robot. All washes and addition of the detection reagents is performed automatically. The plate is then transferred to the SmartReader and results are exported to the LIMS system for reporting.
Clinical Data
The UltraPlex Coeliac assay has been tested side-by-side with a market-leading ELISA assay for anti-Gliadin IgA antibodies and a leading ELISA assay for anti-tTG IgA antibodies. More than 400 samples have been compared in an independent laboratory, and UltraPlex Coeliac yields comparable performance to the competitor products.
